Role of TGF-β and oxidative stress in human pathologies

Transforming growth factor-beta (TGF-β) is a cytokine that regulates tissue homeostasis and is dysregulated in various human pathologies. Our SGR group studies alterations in the signaling pathway of this cytokine, which is overactivated in two large groups of human pathologies: a) Liver cancer, particularly due to its role in invasion and metastasis; b) Cardiovascular genetic diseases related to connective tissue, in particular aortic aneurysm in Marfan syndrome (MFS) and aortic stenosis and heart disease in Williams-Beuren syndrome (WBS).

In recent years, we have progressed in the study of the TGF-β signaling pathway and have identified oxidative stress as a key element in the formation and progression of these pathologies. We are also testing new therapeutic tools to inhibit TGF-β hypersignaling. We want to advance in: i) the role of TGF-β and oxidative stress in the tumor microenvironment; ii) discover new targets related to TGF-β and redox stress in the pathogenesis of both aortopathies.

Objectives

Role of TGF-β, NADPH oxidases and oxidative stress on the liver tumor microenvironment:

1.1 Primary tumors: hepatocellular carcinoma and cholangiocarcinoma.
1.2 Liver metastasis, both intrahepatic and extrahepatic, focused on melanoma.

 

New TGF-β-based targets onto redox stress effectors in the pathogenesis of the aortopathy:

2.1. To identify additional sources of ROS in the progression of the aortopathy in Marfan Syndrome (MFS) and Williams-Beuren syndrome (WBS): focus on xanthine oxidoreductase and role of uric acid as pro-oxidant or antioxidant.
2.2. To apply new pharmacological therapies interfering to redox stress ameliorating the cognitive and cardiovascular injuries occurring in both diseases.
